چکیده

Plasmid encoded protein (Pet), a product of E. coli, infiltrates leukocyte in experimental models and acts on macrophages as a chemokine. As an immune response orchestrator, macrophages play an important role in inducing proinflammatory cytokines. In this work, we tested whether in macrophages Pet stimulates the expression of IL1, TNF , MIF and the antagonist IL1Ra. We also sought the IKK /NF B pathway as a possible mediator. We found that indeed Pet increases the mRNA of inflammatory cytokines and the antagonist IL1Ra with different rates and kinetics through the IKK /NF B pathway, which could be consistent as a factor in chronic inflammation.

Activation of IKK by thymosin alpha1 requires the TRAF6 signalling pathway.

Thymosin alpha1 (T(alpha)1) is noted for its immunomodulatory activities and therapeutic potential in treatment of infectious diseases and cancer. However, the molecular mechanism of its effectiveness is not completely understood. Here, we report that T(alpha)1 induces interleukin (IL)-6 expression through the I(kappa)B kinase (IKK) and nuclear factor-(kappa)B (NF-(kappa)B) pathway. Using IKK(b...

Tumor necrosis factor alpha inhibition of hepatitis B virus replication involves disruption of capsid Integrity through activation of NF-kappaB.

Chronic infection by hepatitis B virus results from an inability to clear the virus, which is associated with liver disease and liver cancer. Tumor necrosis factor alpha (TNF-alpha) is associated with noncytopathic clearance of hepatitis B virus in animal models. Here we demonstrate that the nuclear factor kappaB (NF-kappaB) signaling pathway is a central mediator of inhibition of hepatitis B v...

Effects of anakinra on clinical and radiological outcomes in rheumatoid arthritis.

Interleukin 1 (IL1) plays a central part in the pathophysiology of rheumatoid arthritis (RA). 2 The IL1 gene family includes IL1α, IL1β, and IL1 receptor antagonist (IL1Ra). IL1α and IL1β are both agonist molecules. There are two distinct IL1 receptors, designated type I (IL1RI) and type II (IL1RII). IL1 binding to IL1RI results in signal transduction and cell activation. IL1Ra is the third mem...

Protein kinase C-associated kinase regulates NF-κB activation through inducing IKK activation.

Activation of the transcription factor NF-κB induced by extracellular stimuli requires IKKα and IKKβ kinase activity. How IKKα and IKKβ are activated by various upstream signaling molecules is not fully understood. We previously showed that protein kinase C-associated kinase (PKK, also known as DIK/RIP4), which belongs to the receptor-interacting protein (RIP) kinase family, mediates the B cell...

TNF and IL-1 exhibit distinct ubiquitin requirements for inducing NEMO–IKK supramolecular structures

Nuclear factor κB (NF-κB) essential modulator (NEMO), a regulatory component of the IκB kinase (IKK) complex, controls NF-κB activation through its interaction with ubiquitin chains. We show here that stimulation with interleukin-1 (IL-1) and TNF induces a rapid and transient recruitment of NEMO into punctate structures that are anchored at the cell periphery. These structures are enriched in a...
